Array Analysis
Description:
Comparative analysis for glycoproteins can be performed
using the lectin based GlycoArray Kit (Qiagen, #37702).

2. Labeled streptavidin for biotinylated proteins
3. Labeled glycoprotein primary antibodies

Odyssey Data:
The GlycoArray contains lectins with various glycoprotein specificities (left).Each
slide contains two replicated blocks of lectins. Each block has 12 squares
containing nine spotted lectins in a 3 x 3 pattern. A single lectin is replicated
three times in each square. The kit includes analysis software for determination
of the type of glycoprotein interactions. The analysis gives structural characteristics
for each glycoprotein (i.e. type of N-linked branching). The kit is not made
for quantitative determination of glycoprotein content in sample, but better
suited for comparative analyzes.
The GlycoArray slides were processed using
the Qiagen protocol substituting IRDye labeled conjugates
for visible probes. Purified lactoferrin was added to the
Lactoferrin Positive slide—200 nM lactoferrin. Water was used in the
negative control slide. Rabbit anti-lactoferrin primary antibody
was then added, followed by IRDye® 800CW goat anti-rabbit. The
slides were scanned at 21 μm on an Odyssey® Infrared
Imaging System (left).
