SOUTHERN BLOT

UMC60 Probe Insert

Figure 1. A probe for the UMC60 insert was labeled by PCR incorporation of a biotinylated nucleotide, using 60% Biotin-16-dUTP and 40% dTTP in the reaction. 150ng of probe used, achieving detection of ~3.15 attomoles of insert. Light blue pixels indicate saturation of signal.

Application Overview

Fluorescent Detection of Southern Blots
can be performed with the Odyssey Systems

  • Label probe by incorporation of the desired nucleotide:
    Biotin-16-dUTP* (Roche Applied Science, P/N 11093070910)

    Aminoallyl dUTP*

    • Direct direction after hybridization and washing

  •

  • Sensitivity equivalent to chemiluminescence

    • Fluorescent method allows accurate quantification

    • No hazardous materials or radioactivity

    • No waiting during exposures or film processing

    • Sharp bands with no "blow-out”

* LI-COR Biosciences recommends that all restrictions placed on product labels and product inserts for biotin-16-dUTP and aminoallyl dUTP be followed. Applications other than those recommended on the product inserts may require a license under certain patents owned by third parties. LI-COR Biosciences does not grant any additional license to make, use or sell these products.
