cDNA Sequencing

The combination of Simultaneous Bi-directional Sequencing (SBS™) and the long reads of the LI-COR IR2 System provides the best approach to sequencing full-length cDNA clones.

In bi-directional reactions, standard forward and reverse primers are labeled with different infrared dyes and combined in one reaction. Both ends of cDNA clones are sequenced simultaneously, with read lengths over 1000 bases from each end.

Since the average cDNA (2kb) can be sequenced with one reaction, the time and expense of custom primers is eliminated. The cost savings of producing two sequences per reaction with standard primers, combined with high sensitivity infrared detection also results in very low reaction costs.

Sequencing Reactions

  • Complete coverage of 2kb cDNA inserts with one SBS™ reaction.
  • 1000-1400 bp read lengths from each end of the cDNA insert.
  • Inexpensive - generally requires only two standard vector primers.

Electrophoresis

  • KBPlus Gel Reagents- ready-to-use solution provides consistent results.
  • No post-reaction cleanup.
  • No crosstalk between detection channels ensures high quality data.

Base Calling

  • Adaptive base calling software trains itself while automatically sequencing high resolution, 3-dimensional image data.
  • 99% accuracy at 1000 bases.
