With the Odyssey System, the blotting step can be eliminated by directly detecting proteins inside the polyacrylamide gel. Infrared detection produces clear sharp bands with better sensivity than chemiluminescent, in-gel Westerns.

| Two gels were loaded with purified human Transferrin protein starting at 10ng/per lane, then two-fold dilution. Both gels were processed following the same protocol as Pierce In-Gel western kit, except with the Odyssey system 5% BSA in PBST was used as the antibody dilution buffer. The primary antibody was rabbit anti Transferrin at 1:250 dilution. The secondary antibody was Alexa 680 goat anti rabbit 1:1500 for the Odyssey system, and goat anti rabbit horseradish peroxidase-conjugated antibody at 1:500 dilution for Pierce kit. | |
