Two Color In-Gel Western Analysis

With the Odyssey System, the blotting step can be eliminated by directly detecting proteins inside the polyacrylamide gel. Infrared detection produces clear sharp bands with better sensivity than chemiluminescent, in-gel Westerns.

Chemiluminescent In-Gel Western

Two gels were loaded with purified human Transferrin protein starting at 10ng/per lane, then two-fold dilution. Both gels were processed following the same protocol as Pierce In-Gel western kit, except with the Odyssey system 5% BSA in PBST was used as the antibody dilution buffer. The primary antibody was rabbit anti Transferrin at 1:250 dilution. The secondary antibody was Alexa 680 goat anti rabbit 1:1500 for the Odyssey system, and goat anti rabbit horseradish peroxidase-conjugated antibody at 1:500 dilution for Pierce kit.
