WesternSure ECL Stripping Buffer is ideal for stripping and reprobing chemiluminescent Western blots.

WesternSure ECL Stripping Buffer is not recommended for loading amounts over 30 µg. It will demonstrate decreased performance when used at 2X or 5X concentration and heating the solution will not increase the stripping efficiency.

Original: EGF-stimulated A431 lysate (10 and 5 µg total protein) was probed with mouse anti-ERK2 followed by detection with WesternSure HRP Goat anti-Mouse (LI-COR P/N 926-80010) and WesternSure PREMIUM Chemiluminescent Substrate (LI-COR P/N 926-95000). The blot was exposed to film for 20 sec. Stripped: The blot was then stripped with WesternSure ECL Stripping Buffer (LI-COR P/N 926-92000), followed by detection with WesternSure PREMIUM, and exposed to film for 10 min. Reprobed: The blot was re-blocked and reprobed with Beta-Actin Monoclonal (LI-COR P/N 926-42212) followed by detection with HRP Goat anti-Mouse and WesternSure PREMIUM. The blot was exposed to film for 20 sec.

Human transferrin (400 ng to 12.5 ng/lane) was probed with rabbit anti-transferrin (1:1000) followed by detection with HRP conjugated Goat anti-Rabbit IgG (LI-COR P/N 926-80011) and WesternSure PREMIUM Chemiluminescent Substrate (LI-COR 926-95000). Chemiluminescence was detected with Odyssey® Fc Imaging System using a 2 minute acquisition (Original). The blot was then stripped using WesternSure ECL Stripping Buffer (LI-COR P/N 926-92000) and imaged on Odyssey Fc. (Stripped) The blot was reprobed with different concentrations of the same primary and secondary antibodies (1:5,000 and 1:50,000 respectively) followed by detection with WesternSure PREMIUM.
